After 18 months of storage at -150 C, some larvae of Ditylenchus dipsaci, which had been treated in a 7.5% solution of dimethyl sulphoxide and cooled to -25 C before storage, were still viable on thawing. Some survivors penetrated and developed normally in stems of alfalfa seedlings. Tests showed that active larvae could be frozen directly, thus eliminating the need to use the quiescent stage of this nematode previously thought necessary for successful storage at cryogenic temperatures. The method described is suitable for long-term storage of D. dipsaci and may, with slight modifications, be used to preserve other plant-parasitic nematodes. 相似文献

Three components of the survival strategy of a terrestrial Antarctic mite, Alaskozetes antarcticus (Acari: Cryptostigmata) are considered: overwintering survival, energetics and life history. Supercooling is an important feature of its cold tolerance, whilst elevation of standard metabolism allows activity at low temperatures, both of which contribute tcTa long development and maximum survival of individuals in the population. These are facets of the overall survival strategy evolved by such a species in response to the Antarctic terrestrial environment, but which may be widespread in polar invertebrates. 相似文献

Daily and seasonal fluctuations in temperature present significant challenges for the survival of many ectothermic species that can be tempered via thermal acclimation. In the present study, we use multiple naturally derived genotypes of Drosophila melanogaster to determine the persistence of beneficial short‐term thermal acclimation on subsequent survival after cold shock. We found that the benefit of short‐term acclimation persisted for 2 h in most genotypes after a rapid cold hardening treatment. Genotype did not directly influence the persistence of short‐term acclimation benefits, indicating that environmental variation may be more important for the persistence of acclimation benefits rather than genetic capacity for acclimation. The present study extends the current understanding of the limits and importance of short‐term acclimation events, providing greater detail on the timing of the loss of short‐term acclimation benefits in a genetically variable natural population. 相似文献

Pancreatic cancer has a dismal prognosis and to date there are no targeted therapies for this malignancy. Using shotgun proteomics, the mRNA binding protein cold shock domain containing E1 (CSDE1), also called upstream‐of‐N‐Ras, is detected in pancreatic cancer cell lines but not in normal pancreatic epithelial cells. The expression of CSDE1 in pancreatic cancer cells is confirmed by Western blotting and immunohistochemistry of human pancreatic tumors. In vitro functional assays show that siRNA downregulation of CSDE1 or gene knockout using CRISPR‐Cas9 significantly reduce the invasiveness of pancreatic cancer cells. Together, this study reveals that CSDE1 is overexpressed in pancreatic cancer and is a potential therapeutic target to inhibit pancreatic cancer cell invasion. 相似文献

Extreme weather conditions such as cold stress influence the productivity and survivability of beef cattle raised on pasture. The objective of this study was to identify and evaluate the extent of the impact of genotype by environment interaction due to cold stress on birth weight (BW) and weaning weight (WW) in a composite beef cattle population. The effect of cold stress was modelled as the accumulation of total cold load (TCL) calculated using the Comprehensive Climate Index units, considering three TCL classes defined based on temperature: less than −5°C (TCL5), −15°C (TCL15) and −25°C (TCL25). A total of 4221 and 4217 records for BW and WW, respectively, were used from a composite beef cattle population (50% Red Angus, 25% Charolais and 25% Tarentaise) between 2002 and 2015. For both BW and WW, a univariate model (ignoring cold stress) and a reaction norm model were implemented. As cold load increased, the direct heritability slightly increased in both BW and WW for TCL5 class; however, this heritability remained consistent across the cold load of TCL25 class. In contrast, the maternal heritability of BW was constant with cold load increase in all TCL classes, although a slight increase of maternal heritability was observed for TCL5 and TCL15. The direct and maternal genetic correlation for BW and maternal genetic correlation for WW across different cold loads between all TCL classes were high (r > 0.99), whereas the lowest direct genetic correlations observed for WW were 0.88 for TCL5 and 0.85 for TCL15. The Spearman rank correlation between the estimated breeding value of top bulls (n = 79) using univariate and reaction norm models across TCL classes showed some re-ranking in direct and maternal effects for both BW and WW particularly for TCL5 and TCL15. 以生菜(Lactuca sativa)种子为研究对象,通过不同时间的吸水处理分析其含水量变化,再通过程序降温处理,分析不同含水量种子发芽率的差异,以及脂肪酸合成有关基因(FAD2、FAD3、PPT、ELOVL)和冷调节基因ICE1的表达。结果表明,种子含水量随吸水时间增加而升高。程序降温至同样的低温冷冻条件下(-20℃、-22℃),吸水时间小于6 h的种子发芽率较高,而吸水8 h以上的种子发芽率显著降低。种子吸水8 h含水量处于饱和状态,在此状态下种子对低温较为敏感,说明含水量对种子耐冻性有影响。冷冻处理后生菜种子基因表达检测结果表明,脂肪酸去饱和酶基因(FAD2、FAD3)、蛋白质棕榈酰基硫脂酶相关基因(PPT)、长链脂肪酸延伸酶相关基因(ELOVL)的表达水平均随着种子含水量增加呈上升趋势,吸胀10 h的种子表达量最高,此时种子由于高含水量所受冷冻伤害最大。基因ICE1在冷冻处理种子中的表达也随着吸水时间增加而升高,在吸水10 h时种子中表达量到最高水平。综上,种子含水量越高,所受冷冻伤害越大。但种子在低温条件下具有一定的抗冷反应,可通过相关基因的过表达调控合成更多不饱和脂肪酸、抗冻蛋白等提高含水种子耐冻性。 相似文献
